Background:Dapoxetine is a selective serotonin reuptake inhibitor (SSRI) and a common treatment for premature ejaculation (PE) in men. We recently published our experience with dapoxetine in treating PE and premature ejaculation (PE) in men. In addition, we evaluated the safety and effectiveness of dapoxetine treatment for PE and the safety and tolerability of dapoxetine treatment for the treatment of PE. We hypothesized that dapoxetine treatment of PE would be safe, effective, and well tolerated.
Methods:This prospective, multicenter, randomized, placebo-controlled trial was conducted to evaluate the safety of dapoxetine treatment of PE and the safety of dapoxetine treatment of PE in men with PE. The protocol included baseline, titrated, and follow-up assessments. The study was conducted in three randomized groups: control, dapoxetine, and placebo. All study participants completed a standardized medical questionnaire (TIMS).
Results:Patients with PE or PE (n=928) and those without PE were compared with 928 control and placebo participants (n=859). Baseline demographics and laboratory parameters did not significantly differ between the study groups. The average duration of dapoxetine treatment was 8.4 days, with an average of 5.7 days in the control group and 3.4 days in the dapoxetine group. There were no significant differences between the study groups in the duration of dapoxetine treatment. There were no significant differences in the incidence of adverse events in either group. The mean time to ejaculation was 2.7 hours, with an average of 2.6 hours in the control and 2.2 hours in the dapoxetine group. There was a decrease in the number of premature ejaculation events (p=0.01) and a reduction in the percentage of premature ejaculation events (p=0.01), with a mean difference of -1.4% and a median difference of -2.3% (95% CI, -0.6 to 1.5).
